Project title: Influence of titanium on inflammation in peri-implant tissues

General information about third-cycle studies: The PhD education in Odontological Science corresponds to studies of 240 credits in total. The studies are equivalent to four year full-time and lead to a Degree of Doctor. The studies comprisecarrying out a scientific project and completing at least 30 credits of courses at third-cycle level. Project: Peri-implantitis is a pathological condition occurring in patients with dental implants. The aggressive host response in peri-implantitis may be affected by the specific environment around the dental implant made of titanium. The current series of studies aims at evaluating the influence of titanium on inflammation in peri-implant tissues.

In Study1 the occurrence of titanium particles in peri-implant tissues and the influence of implant characteristics, installation protocols and healing time will be assessed. Archive material from 500 implant units from pre-clinical in vivo experiments will be used.Differences in local host response in peri-implantitis and periodontitis in humans with specific focus on epigenetic changes in neutrophils and other cells of the lesions will be analyzed in Study 2. Soft tissue biopsies from 40 patients with severe peri-implantitis and 40 patients with severe periodontitis will be used. Assessments of damage patterns in neutrophils using the markers for DNA-damage and hydroxyl radical-induced injury will be performed. Study 3 aims to determine if deposition of titanium is more pronounced in inflamed than in non-inflamed peri-implant tissue. Soft tissue biopsies from 20 patients with severe peri-implantitis and healthy peri-implant tissues will be collected. Histological and elemental analysis with specific focus on titanium will be made.In Study 4 the effect on peri-implant tissues of using a mechanical, abrasive decontamination technique during surgical therapy of experimental peri-implantitis will be evaluated. Following experimental peri-implantitis in a pre-clinical model, surgical treatment will be performed using a mechanical or chemical cleaning procedure. The histological analysis will include assessments of presence, size and density of titanium particles. The aggressive nature and rapid progression of peri-implantitis require early detection. Untreated peri-implantitis may not only lead to loss of implants but also loss of prosthesis and function. The findings will provide important information for minimizing risks for peri-implant diseases in patients with dental implants.

To be eligible for third-cycle studies, the applicant must meet both the general and specific entry requirements.

A person meets the general entry requirements for third-cycle studiesif he or she:

1. has been awarded a second-cycle qualification 2. has satisfied the requirements for courses comprising at least 240 credits of which at least 60 credits were awarded in the second-cycle, or 3. has acquired substantially equivalent knowledge in some other way in Sweden or abroad A person meets thespecific entry requirements for third-cycle studiesif he or she: has successfully completed the English B/6 course or is considered to have acquired equivalent knowledge through previous studies

In selecting between applicants who meet the general and specific entry requirements, their ability to benefit from third-cycle studies shall be taken into account. The assessment shall attach particular importance to documented qualifications for:

– specific knowledge and skills within the subject area and related research fields – experience of scientific theory and relevant research methodology – scientific analysis and presentation verified through thesis, degree project, scientific journals or the like. In addition to documented experience from microscopic analysis oftissue samples, a documented experience from biopsy sampling of peri-implant tissues in conjunction with surgical treatment of patients with peri-implantitis is of value.Experienceof a level similar to that of a dentist with a recognized specialist training in the field is desired.Communication with patient and implementation of sampling in connection with treatment is facilitated by adequate Swedish language skills.

Regulations for employment of doctoral students are given in SFS 1998:80.Only those who are admitted to third-cycle studies may be appointed to doctoral studentships. Those appointed to doctoral studentships shall primarily devote themselves to their studies. They may, however, work to a limited extent with educational tasks, research and administration according to the regulations.

The initial appointment may apply for no longer than one year. An appointment may be renewed for no more than two years at a time. The total employment period may not exceed the time corresponding to full-time third-cycle studies for four years.

Salaries for doctoral students are regulated in a local agreement at the University.
