PhD Scholarship Position in Pharmaceutical Science Pharmacokinetic/-dynamic Modelling

The Department of Pharmacology is located at the Institute of Neuroscience and Physiology at the Faculty of Health Care Sciences at the Sahlgrenska Academy, University of Gothenburg. Several research groups are active at the Department and research is conducted within pharmacology, in particular regarding the central and autonomic nervous system, pharmacokinetics and drug metabolism, oncology, addiction and rational use of medicines. We teach at an undergraduate and graduate level at most programmes within the Sahlgrenska Academy, not the least at the Pharmacy and Medical programmes.

Project title: Pharmacokinetic/pharmacodynamic modelling and simulation of the exposure-response and time-course of effects of N,N-dimethyltryptamine (DMT) administered to human beings

General information about third-cycle studies: The PhD education in PharmaceuticalScience corresponds to studies of 240 credits in total. The studies are equivalent to four year full-time and lead to a Degree of Doctor. The studies comprisecarrying out a scientific project and completing at least 30 credits of courses at third-cycle level. Project: N,N-dimethyltryptamine (DMT) is an endogenous serotonergic compound with psychedelic effects. It is currently being investigated as a therapeutic agent in the treatment or amelioration of a number of psychiatric conditions such as post-traumatic stress disorders, depression and end-of-life anxiety. Its disposition in the human body is virtually unknown. This project will for the first time investigate DMT pharmacokinetics in human subjects. Simultaneous mathematical modelling of the time courses of both DMT plasma concentrations and clinical effect variables will seek to elucidate the characteristics of the exposure-response relationships. A sensitive LC-MS/MS method of DMT quantitation in plasma will be validated. Whether active DMT transport occurs across the blood brain barrier will be studied e.g. by CNS microdialysis in laboratory animals. Results will be used to design appropriate dose regimens by mathematical simulation to achieve prolonged exposure and thus enabling improved conditions for neuroimaging by fMRI, MEG and/or EEG.

The PhD student will become a research member of the Unit for Pharmacokinetics and Drug Metabolism (PKDM), Department of Pharmacology, Institute of Neurosciences and Physiology at the Sahlgrenska Academy. The Unit is richly endowned with tandem masspectrometers for drug analyses. This PhD position will entail several international academic collaborations. Our Unit has been described as a globally premiere academic unit for training in pharmacokinetics (Tucker, BJCP, 2012;73:924-6).

To be eligible for third-cycle studies, the applicant must meet both the general and specific entry requirements.

A person meets the general entry requirements for third-cycle studiesif he or she:

1. has been awarded a second-cycle qualification 2. has satisfied the requirements for courses comprising at least 240 credits of which at least 60 credits were awarded in the second-cycle, or 3. has acquired substantially equivalent knowledge in some other way in Sweden or abroad A person meets thespecific entry requirements for third-cycle studiesif he or she: has successfully completed the English B/6 course or is considered to have acquired equivalent knowledge through previous studies

In selecting between applicants who meet the general and specific entry requirements, their ability to benefit from third-cycle studies shall be taken into account. The assessment shall attach particular importance to documented qualifications for:

– specific knowledge and skills within the subject area and related research fields – experience of scientific theory and relevant research methodology – scientific analysis and presentation verified through thesis, degree project, scientific journals or the like. Documented experience in primarily pharmacokinetics and secondly in pharmacodynamics and/or drug metabolism is of specific value.Experience in modelling and simulation of biological systems, preferably using non-linear mixed effects approaches andof drug quantitation in biological matrices using liquid chromatographic methodology is also valuable.Moreover,a high capability to assimilate and construe new knowledge, good communication skills, as well as a high motivational factor are highly desirable.

Regulations for employment of doctoral students are given in SFS 1998:80.Only those who are admitted to third-cycle studies may be appointed to doctoral studentships. Those appointed to doctoral studentships shall primarily devote themselves to their studies. They may, however, work to a limited extent with educational tasks, research and administration according to the regulations.

The initial appointment may apply for no longer than one year. An appointment may be renewed for no more than two years at a time. The total employment period may not exceed the time corresponding to full-time third-cycle studies for four years.

Salaries for doctoral students are regulated in a local agreement at the University.
