PhD Scholarship Position in Medical Science Hormone Secretion in White Adipcotyes

The Department of Physiology is part of the Institute of Neuroscience and Physiology at Sahlgrenska Academy, the Faculty of Health and Care Sciences at the University of Gothenburg. The Department conducts research within the area of physiology. Several projects involve a close collaboration with the Sahlgrenska University Hospital. We offer undergraduate courses as parts of study programs in Medicine, Dentistry, Pharmacy, Speech and language pathology, Dental hygiene, Occupational therapy, Physiotherapy and the bachelor program of Cognitive Science. The research groups at the Department represent a broad range of research areas within the field of physiology.

Project title: Molecular and cellular regulation of white adipocyte adiponectin exocytosis - the role of caveolae and exosomes

General information about third-cycle studies: The PhD education in Medical Science corresponds to studies of 240 credits in total. The studies are equivalent to four year full-time and lead to a Degree of Doctor. The studies comprisecarrying out a scientific project and completing at least 30 credits of courses at third-cycle level. Project: The aim of our research is to understand how secretion of the white adipocyte hormone adiponectin is regulated and how this control may be disturbed in metabolic disease. Adiponectin levels are decreased in obese/type 2 diabetic individuals and high adiponectin levels protect from development of diabetes, thus suggesting adiponectin as a promising pharmacological target for prevention and treatment of metabolic disease. The specific aim of this PhD project is to define the importance of caveolae (plasma membrane lipid raft invaginations) and exosomes (nanovesicles) for adiponectin release. Our hypothesis is that caveolae have an important function for release of adiponectin via an exosomal pathway.

Using a combination of high-resolution fluorescence microscopy studies in live cells and in vivo investigations in mice we will: 1) Define the dynamics and molecular control of adiponectin vesicle exocytosis; 2) Determine the role of caveolae and exosomes for exocytosis of adiponectin. The project also involves several standardised cell and molecular biological/biochemical techniques (cell culture, transfection/transduction, PCR, siRNA, ELISA, Western blot, cloning etc). The PhD student will belong to the section of Metabolic Physiology. Extensive time, especially during the first year, will however be spent in the group of co-supervisor Sebastian Barg, Department of Medical Cell Biology, Uppsala University. I this group, the student will learn advanced fluorescence microscopy techniques, essential to establish the studies in Gothenburg. The PhD position offers an opportunity to conduct scientific research within highly motivated environments, with extensive competence within the topical research field as well fully equipped laboratories. Comprehensive mentoring, opportunities for conference presentations, and further support for career development will be provided. To be eligible for third-cycle studies, the applicant must meet both the general and specific entry requirements.

A person meets the general entry requirements for third-cycle studiesif he or she:

1. has been awarded a second-cycle qualification 2. has satisfied the requirements for courses comprising at least 240 credits of which at least 60 credits were awarded in the second-cycle, or 3. has acquired substantially equivalent knowledge in some other way in Sweden or abroad A person meets thespecific entry requirements for third-cycle studiesif he or she: has successfully completed the English B/6 course or is considered to have acquired equivalent knowledge through previous studies

In selecting between applicants who meet the general and specific entry requirements, their ability to benefit from third-cycle studies shall be taken into account. The assessment shall attach particular importance to documented qualifications for:

– specific knowledge and skills within the subject area and related research fields – experience of scientific theory and relevant research methodology – scientific analysis and presentation verified through thesis, degree project, scientific journals or the like. Documented experience in the field of metabolic health and disease, specifically with focus on regulation of hormone secretion and/or adipose tissue functionality, as well as in fluorescence microcopy techniques is of value. Good communication skills, and a very high level of commitment and curiosity about research in the current field are desirable.

Regulations for employment of doctoral students are given in SFS 1998:80.Only those who are admitted to third-cycle studies may be appointed to doctoral studentships. Those appointed to doctoral studentships shall primarily devote themselves to their studies. They may, however, work to a limited extent with educational tasks, research and administration according to the regulations.

The initial appointment may apply for no longer than one year. An appointment may be renewed for no more than two years at a time. The total employment period may not exceed the time corresponding to full-time third-cycle studies for four years.

Salaries for doctoral students are regulated in a local agreement at the University.
